Martin Becker and colleagues developed an enzyme cascade for the synthesis of 2′3′-cGAMP from nucleosides.

Enzyme cascades enable multi-stage syntheses in one-pot reactions without the need to purify intermediates. First, the substrate spectrum of nucleoside kinases and polyphosphate kinases was investigated. Based on the results, an enzyme cascade for 2′3′-cGAMP synthesis from guanosine was developed. Subsequently, the cascade was extended to enable the first 2′3′-cGAMP synthesis from the nucleosides adenosine and guanosine in four reaction steps. In total, 57 % of guanosine was converted to 2′3′-cGAMP. This shows that it is possible to develop shorter synthesis routes for drugs by combining several biocatalytic reactions, thus supporting the chemical and pharmaceutical industry's goal to move towards more sustainable processes.
